Abstract
DEVELOPMENT OF URINE SAMPLE PREPARATION METHOD USING DISPERSION SOLID PHASE EXTRACTION BASED ON ACTIVATED CARBON FOR THE ANALYSIS OF CREATININ IN SPECTROPHOTOMETRY Rizki Nur Hidayat Email : [email protected] ABSTRACT Creatinine is a chemical compound called 2-amino-1-methyl-2-imidazole4-on, which is the end result of skeletal muscle metabolism. Creatinine levels in the blood will increase when kidney function decreases. The decreasing of kidney performance also effect of decreasing creatinine level in urine. Sample preparation method based on dispersive solid phase extraction using carbon active to the determinastion of creatinine in spectrometry was successfully developed. Extraction process was performed by dispersing 5 gr of activated carbon in sample solution containing creatinine 5 ppm and then stired using magnetic stirrer for 30 minutes. In the end of extraction process, activated carbon and sample solution was separated, then activated carbon transferred into sentrifuge tube and added with 5 mL of ethanol for desorption process using vortex. After desorption process, the filtrate was analyzed using spectrophotometer UV-Vis in wavelength 205 nm. Several of optimation parameters was investigated and showed that optimum mass adsorbent for 0,5 gr and optimum extraction time for 30 minutes. Under optimum extraction condition, the extraction technique showed a good linearity in the range concentration of 1-5 ppm with linearity value 0,9954. The percentage recoveries were generally achieved in the range of 95-103% with the precission of 4-14%. This method was also successfully applied to the determination of creatinine in urine sample with good percentage recovery of 112% and perecission of 0,45%.
