Abstract
Inhibition Test of Formation of Escherichia coli Biofilm Producing Extended Spectrum Beta Lactamases (ESBLs) from Nanoemulsion of Galangal Oil (Alpinia galanga [L] Willd) Ike Yuyun Winarsih Email : [email protected] ABSTRACT Escherichia coli is a highest Extended-Spectrum Beta- Lactamase (ESBL) producing bacteria and becomes a biofilm producer. Biofilm is a collection of complex microbes wrapped in matrices Extracellular Polymeric Substances (EPS) which develop when a group of microorganisms is irreversibly attached to inert surfaces, metals, plastics, mineral surfaces, even living tissues including humans. Biofilms cause bacteria to be resistant to extreme conditions, antibiotic and disinfectant. This has become a major threat in the fields of health, food industry, the drug industry and so on. One of the methods for handling biofilms using essential oil in nanoemulsion formulations as antibiofilm agents. The aim of this research was to determine the antibiofilm activity of galangal oil nanoemulsion (Alpinia galanga [L] Willd) on the formation of E.coli biofilm in vitro, prevention, inhibition and degradation of biofilms. This research uses a Microtitter Plate Biofilm Assay method to grow biofilms and the testing of galangal oil nanoemulsion. Measurement of biofilm formation by microplate reader and quantitative data is obtained in the form of Optical Density (OD) with a wavelength of 595 nm. The results showed that at a concentration of 22.5 mg / mL the given galangal oil nanoemulsion could provide a percentage inhibitory value for E.coli biofilm formation producing ESBLs up to 58% at 24 hours incubation and up to 71% at 48 hours incubation.
